WP3

WP3: Deciphering the role of the gut microbiota from AD patients in the modulation of the metabolite(s) level

We hypothesize that alterations of the gut microbiota composition in AD patients will influence metabolite(s) levels and concomitantly the behaviour in a model of fecal microbiota transplantation (FMT). Our primary objective is to prove the origin of the selected metabolite(s): microbial, co-metabolite or from the host. We will test whether gut dysbiosis seen in AD patients is directly (microbial or co- metabolite) or indirectly (host metabolite) associated with metabolite(s) levels and with behavioural and neurobiological outcomes (secondary objective). Overall, these evaluations will allow the consortium to confirm the origin of the metabolite and to formulate mechanistic hypothesis. It will lay the foundation of the dissection of molecular mechanism through which the identified metabolite(s) modulate brain cells function leading to behavioural alterations (WP4).

Task 3.1 Testing the causal role of gut microbiota on the modulation of metabolite composition.

Task 3.2 Testing the relationship between metabolite levels and behavioural and neurobiological alterations.
